Amorphous Solid Dispersion Screening
With an increasing number of poorly soluble or difficult to crystallize drugs entering development, new formulation strategies are required to achieve sufficient bioavailability. Amorphous solid dispersions (ASDs) are an interesting option receiving increased attention in recent years. ASDs offer the advantages of improved dissolution rates and higher apparent solubility characteristics of amorphous drug compounds.
A successful ASD stabilizes the amorphous state of the drug and provides sufficient resistance to chemical degradation. A variety of factors contribute to the formation of a suitable ASD, including the nature of the polymer, the drug polymer ratio, the impact of surfactants and the solvent used in the process. Finding the optimal combination of these factors is essential.
Avantium has further developed high-throughput solid dispersion screening technology. Recipes for creating ASDs of a compound are screened efficiently and effectively and the results are analyzed using a variety of techniques including our proprietary high-throughput XRPD technology. ASD screening uses small amounts of sample and delivers results in only weeks.
